Using molecular markers for the study of wild basidiomycete fungi

Abstract

Molecular marker techniques in the study of wild basidiomycetes are increasingly applied to ecology projects, with a special focus on genetic diversity analysis. Specialized methods are often required for the extraction of DNA from organisms in natural environments, due to the complex compounds that constitute them (carbohydrate polymers) and environmental contaminants (soil particles). The biological materials used were basidiocarps collected in the forest of Santa Rosa, Gto., and mycelium isolated from these basidiocarps. In this work, a DNA extraction method was used that allowed PCR amplification, restriction enzyme digestion, and Southern hybridization by a non-radioactive method. The results obtained: Amplification of the ITS1 region of the ribosomal unit of the different species of basidiomycetes. It was possible to observe the genetic diversity between the different species of basidiomycetes and the mycelia. In addition, the results also suggest differences in DNA methylation between the vegetative mycelium and the basidiocarp mycelium. Finally, it is worth mentioning that no previous works were found on the application of non-radioactive Southern hybridization methods for the analysis of wild basidiomycetes, this work being a pioneer in applying this technique.
